Can you please explain some signifances of wearing a brahmana thread. We know it is given with the gayatri mantra initiation.

The sacred thread is given in a ceremony called upanayanam. It is given to "twice-born" persons in Vedic civilization. The second birth is a ritual ceremony or samskara performed between the ages of 8-16. Brahmanas, ksatriyas, and vaisyas all receive the thread. These three classes of person in Vedic civilization perform yajnas and deity worship.

Originally the thread was only worn during mantra chanting, but later on it was worn all the time.

In modern times its in flux as we reestablish Vedic varnasrama society. Presently we think of it as a "brahmana thread", but it is actually a "sacred thread" and is worn by brahmanas and non-brahmanas alike who have undergone the second birth samskara.
